Where exactly do I meet the guide?

At the 25hours Hotel on Pilestræde 65, next to the Round Tower (Trinitatis Kirke). Don't enter the hotel. Wait outside. The staff won't know about your tour.

No. Your skip-the-line tickets give you a reserved time slot, so you enter without the ticket-office queue. You may wait briefly for ticket validation and security checks, but not for entry itself.
