You'll walk past the Sanctuary of St. Catherine, where she actually lived with her large family in a house that's now split into three small churches, then move toward Piazza del Campo — the main square where the Palio takes place and where the Palazzo Pubblico sits like the city's beating heart. Your guide will explain why this race matters so much to the Sienese, and if you're interested, you can visit a Contrada Museum to see the prizes won over centuries. The Baptistery comes next, unfinished at the top (your guide will tell you why the builders stopped), and then the Cathedral itself, which was supposed to become the largest in the world before the project got abandoned. If you want to step inside and see works by Michelangelo, Donatello and Bernini, you can buy tickets on the spot — though note that Sunday mornings the Cathedral doesn't open until 1pm.
